Abstract: Knee osteoarthritis (KOA) is a prevalent degenerative joint disease, and it is a key research objective that its pathogenesis and the development of novel therapeutic interventions are elucidated. The purinergic receptor P2X ligand-gated ion channels play crucial roles in diverse cellular processes, making them potential drug targets based on the pathological mechanisms. As to the pathogenesis of KOA, this paper reviews focuses on the involvement of three specific subtypes of P2X ion channels, namely P2X3, P2X4, and P2X7. Moreover, it aims to explore innovative drug development strategies targeting these P2X ion channels, and to provide reference basis for the mechanism, diagnosis and treatment of KOA.

Key words: Knee osteoarthritis; P2X receptors; Ion channels
