Abstract: Oxidative stress plays a critical role in age-related eye diseases by damaging lens epithelial cells (LECs) and retinal pigment epithelial (RPE) cells. This damage can lead to cellular senescence and apoptosis, ultimately resulting in age-related cataract (ARC) and age-related macular degeneration (AMD). Klotho proteins have garnered significant attention due to their multiple functions, including anti-apoptosis, anti-aging, and antioxidative stress effects. Numerous studies have demonstrated that Klotho proteins exert their antioxidative stress effects primarily by regulating key targets such as FOXO, Nrf2, and NOX. Experimental data further confirm that Klotho proteins can significantly attenuate apoptosis in LECs and RPE cells by inhibiting oxidative stress, thereby potentially reducing the risk of ARC and AMD. The present literature will explore the mechanism of action of Klotho proteins in countering oxidative stress in age-related eye diseases.
