Abstract: Piezo1 is a mechanosensitive ion channel that plays a crucial role in the physiology and pathology of the central nervous system (CNS). Studies have shown that Piezo1 channels regulate the activity of microglia and astrocytes by sensing mechanical force changes, thereby participating in the development and progression of diseases such as Alzheimer’s disease, glioblastoma, and central inflammation. Moreover, Piezo1 influences central lymphatic drainage, offering a new perspective for disease treatment. However, the mechanisms of Piezo1 are complex and challenging to integrate into a unified hypothesis. Future research should further explore its mechanisms and develop specific modulators to provide novel strategies for CNS disease treatment.
